In Vitro Cultivation of Gametocytes of Eimeria Tenella Isolated from Broilers in Jos, Plateau, Nigeria

Abstract

Sporozoites from a characterized Eimeria tenella sporulated oocyst were used for the study. Nine day old embryonic eggs of chicken had their chario-allantric membrane (CAM) inoculated with 0.1ml (104) sporozorites along with penicillin (2000IU) and streptomycin (0.05mg) and incubated at 37oC and 70% humidity. Gametocytes were harvested from the cultures on day 5th – 7th post inoculation from the CAM of dead embryos and concentrarted by centrifigution and washed twice with and stored in phosphate buffer saline (PBS) at 4oC.The gametocytes images from the CAM of the embryonic eggs have similar characteristic structure with that of the ceacum of the broilers.
